Assay: Assessment of Chemosensitivity Testing Evaluation of a Tetrazolium-based Semiautomated Colorimetric
نویسندگان
چکیده
Drug sensitivity assays were performed using a variation of a colori metrie [3-{4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT)1 assay on V79, CHO-AuxBl, CH"C5, NCI-H460, and NCIH249 cell lines following optimization of experimental conditions for each cell line. Results from this assay were compared with data assimi lated simultaneously by clonogenic assay and by dye exclusion assay. Good correlation was observed using the CHO-AuxBl cell line and the pleiotropic drug-resistant mutant CH"C5, with similar degrees of relative resistance observed with both the M l'I and clonogenic assays. Good correlation was observed between the clonogenic and M l'I assays for Ih drug exposures, although the MIT assay was more sensitive to vinblastine. In general, the clonogenic assay was more sensitive when continuous drug exposures were utilized, although this was primarily related to the increased drug exposure time. While the use of the MIT assay in drug sensitivity testing of primary tumor samples is limited, since contaminating normal cells may also reduce the tetrazolium, the M I'I assay can be semiautomated, and therefore it offers a valid, simple method of assessing Chemosensitivity in established cell lines.

متن کاملChemosensitivity testing of human colorectal carcinoma cell lines using a tetrazolium-based colorimetric assay.
The in vitro chemosensitivity of 11 human colorectal cell lines to seven chemotherapeutic agents was determined using a semiautomated tetrazolium-based colorimetric assay (MTT assay). Four of the cell lines were from primary tumors and seven from metastases. Eight lines were from patients with no prior chemotherapy. From assay results, we predict 5-fluorouracil (5-FU) to be the sole active agen...
متن کاملEvaluation of chemosensitivity testing with highly purified tumor cells in 435 patients with gastric carcinoma using an MTT assay.
BACKGROUND We evaluated the results of chemosensitivity testing for gastric cancer using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay in terms of the correlation of chemosensitivity and clinicopathological findings. PATIENTS AND METHODS We analyzed 435 consecutive patients with gastric cancer treated between January 1991 and January 2002. Highly purified fresh h...

The tetrazolium dye, 3-(4,5-dimethylthia/ol-2-yl)-2,5-diphi-iiyltclrazolium bromide (MTT), is reduced by live but not dead cells, and this reaction is used as the end point in a rapid drug-screening assay. It can also be used for accurate determinations of drug sensitivity but only if a quantitative relationship is established between cell number and M l'I forma/an production. We have shown tha...
متن کاملIn vitro Evaluation of Anti-Leishmanial Activities of Zataria Multiflora Boiss, Peganum Harmala and Myrtus Communis by Colorimetric Assay
Background & Aims: Plant extracts and plant-derived compounds are valuable sources of new medicinal agents that are commonly used to treat infectious diseases. The purpose of this study was to evaluate, antiLeishmanial activity of three plant extracts on L.major promastigotes by colorimetric assay as compared to a trivalent antimonial compound (tartar emetic). Methods: Leishmania major promasti...
